10 ppm Criterion Based MACO Calculator
- Purpose: This calculator determines the Maximum Allowable Carryover (MACO) using the traditional 10 ppm criterion. The 10 ppm–based method assumes a fixed maximum allowable concentration of 10 mg per kg (10 ppm) of Active Pharmaceutical Ingredient (API) from the previous product in the next product batch. This Calculator is primarily intended for educational purposes, legacy comparison with historical cleaning validation practices, or preliminary assessment. Regulatory Note: Current regulatory guidelines recommend using Health-Based Exposure Limits (HBEL) i.e. Permitted Daily Exposure (PDE) or Acceptable Daily Exposure (ADE) to establish cleaning limits. This approach is scientifically justified and based on toxicological safety data to ensure patient protection.
